Öğe Antibacterial Characteristics of Nanofiber Structures Obtained by Benzalkonium Chloride Additive Poly (Vinyl Alcohol)/Gelatin(2019) Yolay, Onur; Yüksek, Metin; Tezcan, Erdem; İşgören, Erkan; Saltık, DeryaPolyvinyl alcohol (PVA) is a hydrophilous and semi-crystallized. It has attracted much relevancy due to its pretty chemical endurance, finethermal determination, decent physical specialities, wonderful bio-compatibility and cheapness. Gelatin (G) is a natural polymer and thatare interesting materials for biomedical applications. Electrospinning is a simple method that provides very porous nanofiber productionwith high surface area. It is possible to produce biomedical, filtration, energy storage and protective materials by using electrospinningmethod. Benzalkonium chloride (BAC) is a kind of anti-microbial cationic surface-active agent, which has been pretty used in merchantwound dressings and has a powerful status toward Gr+ bacteria.In this study, nanofibers were produced from electrospinning of BAC, PVA and G containing solutions at various concentrations. The fibersof obtained nanofiber structure were uniform, continuous and intensive. The optimum parameters in terms of good mechanical and antibacterialproperties were determined. S2 sample, electrospun from 11.63% PVA + 1.0% G +1.0% BAC containing solution, had the bestmorphological and mechanical properties due to having the thinnest fiber diameter (51±13nm) and the highest vertical strength (4.299MPa)and horizontal strength (4.058MPa). It also had antibacterial activity against all the bacteria tested (E. coli, P. aeruginosa, B. subtilis andS. aureus). Due to owning good mechanical and antibacterial properties, S2 sample can have many uses in medical sector.Öğe Antibacterial properties of cotton fabrics treated with 8-hydroxyquinoline and borax(CBU Research Inst S R O, 2018) Büyükakıncı, Banu Yeşim; Tezcan, Erdem8-Hydroxyquinoline (8HQ) is a metal chelating agent that also has antibacterial properties against several different bacterial specie, but its use in cotton textile fabrics has not been tested before. 8HQ may be an alternative to heavy metal-based applications for antibacterial textile fabrics therefore, the aim of this research paper was to investigate the antibacterial properties of 8HQ and borax (BX) treated cotton fabrics for the medical textile industry. The homogenous distribution of the treated chemicals was confirmed with SEM/EDX imaging. The antibacterial activities of the fabrics were also tested against Escherichia coli and Staphylococcus aureus bacteria by mimicking the ISO 20645:2004 Standard Test Method. The highest antibacterial property was observed with the 8HQ+BX (8HB) treated cotton samples. Therefore, the 8HB treated cotton products have a potential for use especially in the medical fields mainly due to the absence of heavy metals and bacterial growth.Öğe Antibacterial properties of wool fabrics treated with 8-hydroxyquinoline and boron compounds(Inst. Pielarie, 2020) Tezcan, Erdem ; Büyükakıncı, Banu YeşimIn this study, antibacterial properties of wool fabrics treated with 8-hydroxyquinoline (8HQ) and boron compounds were investigated against E. coil, B. subtilis, S. aureus and P. aeruginosa. Experimental group consisted of wool fabrics treated with 8HQ, boric acid (BA) and borax (BX) at 10 g/L concentration for each using the exhaust process. Control group consisted of untreated and silver nitrate treated wool fabrics. Treatments were performed using meta- and premordanting methods and homogenous distribution of treatment chemicals were confirmed with SEM and EDX images. Wool samples treated with a solution containing 8HQ+BX (10 g/L each) and 8HQ+BA+BX (10 g/L each) showed the highest level of antibacterial activity. Antibacterial activity decreased analogous with the decrease in 8HQ concentration however it was still present at the lowest concentration of 1 g/L for E. coli, B. subtilis and S. aureus. But at least 5g/L concentration of 8HQ was required for antibacterial activity against all four tested bacteria. Antibacterial properties decreased after 5 cycles of washing but did not completely disappear. These antibacterial wool products look promising for the medical fields because of their strong effects against bacteria which grow in wounds and cause nosocomial infections. Bağırsak, kendi hücre sayımızın yaklaşık 10 katı kadar sayıda mikroorganizmaya ev sahipliği yapan önemli bir organımızdır. Bağırsak florası gut flora doğumdan itibaren oluşmaya başlar ve 1 sene içerisinde erişkinlerinkine benzer. Bağırsak florasının bakteri ve mikroorganizma kültürü laboratuvar ortamında henüz simüle edilemediğinden çalışmalar daha çok hastalıklı ve sağlıklı bireylerin bağırsak florasının karşılaştırılması yoluyla yapılmaktadır. Parmakizi gibi bireyden bireye değişen bağırsak florasının bağışıklık sistemi ile yakından ilişkisi vardır. Bağırsak florasının, obeziteden sedefe, Crohn hastalığından ülseratif kolite kadar birçok sayıda ve çeşitte hastalıkla ilişkisi olduğu bulunmuştur. Yapılan deneylerde, cilt hasarı sonrası bakteriden arındırılmış farelerde ve antibiyotik verilen farelerde sedef oluşumu gözlemlenirken, bağırsak florasına müdahale edilmemiş farelerin sedefe çok daha dirençli olması, bağırsak florası ile sedef arasındaki ilişkiyi ortaya koymaktadır. Bağırsak florası ve sedef arasındaki ilişkinin daha da aydınlatılması, sedefin tedavisi veya önlenmesi açısından büyük umutlar vadetmektedir.Öğe Effect of DNA concentration on band intensity and resolution in agarose gel electrophoresis(Van Yüzüncü Yıl University, 2021) Arslan, Mevlut; Tezcan, Erdem; Camcı, Haluk; Avcı, Murat KemalObjective: Agarose gel electrophoresis (AGE) is a widely used method for separating, identifying, and purifying nucleic acids. It has been used intensively in molecular researches and biotechnological applications. In AGE, band resolution and band quality are important parameters for researchers. Until now, many factors such as comb thickness, gel concentration, voltage and buffers have been reported to influence resolution and band quality in AGE. However, effect of DNA concentration on band resolution and intensity in AGE is unclear. Therefore, aim of the study was to investigate the effect of DNA concentration on AGE results in detail. Materials and Methods: Different concentrations of DNA Marker and specific DNA fragments obtained by PCR were analyzed, but the loaded total DNA quantity was not changed. Furthermore, the effect of DNA concentration on AGE was also investigated by designed gel combs with different thicknesses and wideness. Findings: It was shown that DNA concentration did not affect gel resolution, and the effect was not changed by comb thickness and wideness. Also, 2-fold dilution did not affect band intensities while 8-fold dilution significantly affected band intensities of all tested DNA fragments. Conclusion: Concentrating samples, at least 8-fold, and using thin and narrow gel combs can be used for desired results in AGE.Öğe Gıdalarda sıklıkla kullanılan doğal ve yapay tatlandırıcıların sağlık üzerine etkileri(İstanbul Gedik Üniversitesi, 2019) Budak, Gözde; Tezcan, ErdemGıda endüstrisinde kullanılan iki tip tatlandırıcı mevcuttur bunlar bitki kaynaklı doğal tatlandırıcılar ve sentetik tatlandırıcılardır. Doğal tatlandırıcılar, diyabetik hastalar için yararlı maddelerdir. Bu derleme gıda sektöründe en çok kullanılan doğal ve yapay tatlandırıcıların sağlık üzerine etkilerinin gözden geçirilmesini amaçlamaktadır.Öğe Multiple non-essential transition metals are accumulated in carotid atherosclerotic plaques: missing link in atherosclerosis?(Humana Press Inc, 2019) Olcay, Ayhan; Tezcan, Erdem; Cantürk, Emir; İnan, Bekir; Karaoğlu, Hasan; Küçük, Ceyhun; Akdemir, BarışPopulation studies revealed that metal exposure through food, environment, and smoking was related with increased risk of cardiovascular disease. In this study, we determined complex chemical elements in surgical carotid endarterectomy specimens and carotid tissues from autopsies without atherosclerosis. Atherosclerotic plaques from 41 endarterectomies and normal carotid tissue from 30 autopsies were collected and elemental composition was determined by inductively coupled plasma optical emission spectrometry (ICP-OES) method. Eleven (26.8%) patients never smoked in carotid endarterectomy group. One patient was brass souvenir worker and one was goldsmith and others did not have direct contact with metals in the carotid endarterectomy group. Na, Cu, Mn, Bi, Co, Mo, Ni, Pb, Sb, Se, Sn, Ti, and W levels were not different between two groups. Bi, Co, Mo, Pb, Ti, and W were below the detection limit of ICP-OES in both groups. Concentrations of Mg, K, Ca, P, Fe, B, Zn, Al, As, Cr, Pt, and Hg were significantly higher in carotid endarterectomies than normal carotid tissue samples. Cd and S values were significantly higher in autopsy samples. There is significant multiple non-essential transition metal accumulation in atherosclerotic carotid endarterectomy plaques. The cardiovascular consequences of metal toxicity have not been researched adequately due to large emphasis on the role of cholesterol in atherosclerosis.
